丹参对糖尿病肾病大鼠MMP-2,TIMP-1,TGF-β1和IV-C表达的影响 被引量:16

Effect of Salvia miltiorrhiza on the Expression of on MMP-2,TIMP-1,TGF-β_1 and IV-C of Rats with Dibetic Nephropathy

摘要 目的探讨丹参对MMP-2,TIMP-1,TGF-β1和IV-C在糖尿病大鼠肾脏中表达的调节及其与糖尿病肾脏病变的关系。方法SD大鼠随机分成正常组、模型组、对照组和观察组,每组10只。用高糖高脂饲料刺激、加小剂量STZ诱导2型糖尿病大鼠模型,于第16周末处死大鼠,免疫组织化学的方法检测各组大鼠肾组织中MMP-2,TIMP-1,TGF-β1和IV-C的表达,RT-PCR法检测各组大鼠MMP-2,TIMP-1mRNA的表达,同时检测血糖、尿蛋白及肾功能等。结果对照组、观察组与模型组比较,TGF-β1,TIMP-1和IV-C的表达显著减少(P<0.05),MMP-2的表达显著增高(P<0.05),MMP-2,TIMP-1mRNA的表达与组化结果一致。同时,MMP-2及IV-C表达的改变分别与尿蛋白排泄率升高有显著相关性(P<0.05)。结论肾组织中的MMP-2,TIMP-1,TGF-β1和IV-C的表达改变与糖尿病肾脏病变有关,丹参通过影响上述指标的变化对DN的治疗产生作用。 Objective To study the effect of Salvia miltiorrhiza on the expression of MMP- 2 ,TIMP- 1 ,TGF- 131 and IV- C in Kidney of rats with diabetes and its relationship with diabetic nephropathy (DN). Methods SD rats were randomly divided into normal control group, model group, treatment group and observation group, with 10 rats in each group. The type II diabetic rat model was constructed by employing high glucose and lipid, and low dose STZ, and the animals were sacrificed at the end of 16th week. Immunohistochemistry was adopted to detect the expression of MMP -2,TIMP - 1 ,TGF - β1 and IV - C in renal tissues, meanwhile, serum glucose, urine protein and renal function were also evaluated. Results Compared with the treatment group and observation group, the expression of TGF- β1 ,TIMP- 1 and IV- C significantly decreased( P 〈0.05 ), but MMP- 2 expression increased(P 〈 0.05). At the same time, the changes of MMP-2 and IV -C expression were significantly correlated with urine protein excretion. Conclusion The expression of TIMP - 1, TGF - β1 and IV - C are related to DN, Salvia miltiorrhiza has protective effect on DN through affecting the changes of above mentioned indexes.
