IL-6基因转导对人乳腺癌细胞系MCF-7细胞体内外生长特性的影响

摘要 为探讨IL-6基因转导对人乳腺癌细胞体内外生长特性的影响,我们构建了含人IL-6cDNA的逆转录病毒载体pLIL6SN。采用脂质体介导的方法将pLIL6SN导入MCF-7细胞中,对基因转导细胞的体外生长、粘附特性及裸鼠体内的实验性转移能力进行了测定。实验结果表明,经IL-6基因修饰的MCF-7细胞虽显示了明显的体外生长抑制,裸鼠体内实验性转移能力却获得增强。 Objective: To investigate the influence of IL6 gene transduction on human mammary carcinoma cell growth both in vitro and in vivo.Method: retroviral vector pLIL6SN which contained human IL6 cDNA was constructed  and transferred into MCF7 cell line with the mediation of lipofection.IL6 gene transfectant growth and adhesion potential in vitro was assayed and its metastatic competence in nude mice was examined. The results showed that the growth of IL6 genetransferred MCF7 cell was markedly inhibited in vitro,and that the metastatic potential in vivo was enhanced.
