SD大鼠近端肾小管上皮细胞的原代培养及鉴定 被引量:4

Primary culture and identification of proximal renal tubular epithelial cells in SD rat

摘要 采用机械研磨、筛网过滤和酶消化相结合的方法获取肾小管节段进行原代培养,以免疫组织化学法和细胞化学染色法鉴定培养细胞。结果表明,肾小管上皮细胞围绕肾小管节段呈岛屿状向四周逐渐生长并于第4~7天处于对数生长期,培养细胞角蛋白ck18表达阳性,碱性磷酸酶染色阳性,证实培养细胞为近端肾小管上皮细胞,且纯度较高。 The renal tubular segments were collected by mechanical grinding, filtering and chemical digestive methods and the renal tubular epithelial cells were primarily cultured. Immunohistochemical method and cytochemical staining were applied to identify the cell types. The results indicated that renal tubular epithelial cells could grow up to the vicinity gradually around the anchored renal tubular segments, displaying the typical islands layout. They grew in logarithm growing phase from 4 to 7 days. The expression of cytokeratinl8 and alkaline phosphatase(ALP)staining of the cultured cells were positive. Thereby the cultured cells were proved to be proximal renal tubular epithelial cells and its purity was very high.
