摘要
本文介绍一种在RNA聚合酶Ⅱ作用下的体外转录系统,并着重强调该系统成功的关键在于如何制备含有全部转录因子并具有较高的生物活性的全细胞提取液或核提取液。
It is discribed that a procedure for preparing extracts from whole cell or nuclei of human tissue culture cells(Hela), which can be used for determination accurate trancription initiation in vitro from class Ⅱ RNA polymerase. Condition of extraction and assay have been selected for maximum activity, particularly in this paper focused on Soluble cell-free systems which mediate the accurate transcription of eukaryotic cell gene, by RNA-polymerase Ⅱ have been discussed.
