摘要
本文比较详细介绍了DNaseI足纹法(DNase I-footprinting)的原理、基本操作和注意问题,并对其他酶和化学试剂的足纹法及其可能的应用进行了简要讨论。
In this paper, an important technique of footprinting assay to study in gene regulation has been illustrated. The basic principle of general footprinting assay and its procedure in detail has besn indicated. Also, several different kind of footprinting techniques including enzymatic footprinting (DNase I-footprinting, exonuclease in-footprint ing); chemical foot print ing (hydroxyl radical footprinting, OP/Cu-footprinting) and photofootprinting were discussed.We do believe that technique will be used wildly to investigate in eukaryotic gene expression and regulation in the near future.
