摘要
本文报道了一种简易的肝细胞质膜制备方法。肝匀浆经低速离心得到粗制的核与质膜沉淀,然后,使用葡聚糖和聚乙二醇组成的两相分配液再将质膜分离并进一步纯化。电镜和标志酶活性鉴定结果表明,本法能够制备具有一定产率和纯度的肝细胞质膜样品。此外,对本法在肝脏病理研究中的应用价值进行了初步的评价。
A simple method is described in this paper for the isolation and purification of plasma membrane from the rat liver homogenate. The density gradient ultracentrifugation in Neville's method was replaced by an aqueous two phase polymer system consisting of Dextran T500 and PEG 6000 in our method. The plasma membrane has been identified by the electron microscopy and plasma membrane marker enzymes. Furthermore, the method was evaluated in the application to studies of liver pathology.We consider that our method is practical, effective and simple. Whole process only requires less than 5 hours. It is especially suitable to some clinical laboratories.
