摘要
目的:从生物物理和生物化学的角度,探讨由不同比例黄连和吴茱萸组成的汤剂的生物热动力学。方法:黄连和吴茱萸在左金丸中的比例为6:1;甘露散为2:1;茱萸丸为1:1;反左金丸为1:6。利用微量热法,测定了金黄色葡萄球菌在左金丸、甘露散、茱萸丸和反左金丸水煮液作用下的生长热谱曲线,获得相应的生物热动力学参数,结合本草文献报道,综合分析左金丸及类方的药性差异。结果:左金丸及类方的水提取液对金黄色葡萄球菌的生长代谢表现出不同的活性作用。寒凉药方左金丸使细菌指数生长期的生长速率常数显著减小,发热功率显著降低。温热药方反左金丸使细菌指数生长期的生长速率常数和生长代谢过程中热量释放的减少不显著。寒热性能介于二方之间的甘露散和茱萸丸对细菌指数生长期的生长速率常数和热量释放的影响较小。四方之间存在较明显的药性差异,其抑菌作用强弱为:左金丸>甘露散>茱萸丸>反左金丸。结论:微量热法可作为刻画中医经典类方药性差异的有效方法。
AIM : To probe the biothermokinetic property of decoctions made up of a different proportion of Rhizoma coptidis and Fructus evodiae. METHODS: The proportion of Rhizoma coptidis and Fructus evodiae were 6: 1 in zuojin Pill; 2:1 in Ganlu powder; 1:1 in zhuyu Pill and 1:6 in anti-zuojin Pill. Microcalorimetry was applied these Pills and the powder to ohtain the metabolic growth power-time curves of Staphylococcus aureus and biothermokinetic parameters. Comhined with the records of ancient herbal literatures, the drug property difference in Zuojin Pill, Ganlu powder, Zhuvu Pill and antizuojin Pill were synthetically evaluated. RESULTS: The decoctions of Zuojin Pill and its similar prescriptions showed different activities on the growth and metabolism of Staphylococcus aureus in different levels. Drugs, such as Zuojin Pill, with cool property could decrease markedly in the growth rate constant and heat output, while drugs, such as antizuojin Pill, with warm property could decrease poorly in the growth rate constant and heat output. And drugs, such as Ganlu powder and Zhuyu Pill with warm property had poor influence on the growth rate constants and heat output. There were evident drug property differences between them. The sequence of antibacterial activity of the four prescriptions was : Zuojin Pill 〉 Ganlu powder 〉 Zhuyu Pill 〉 Antizuojin Pill. CONCLUSION : Microcalorimetry is a new and useful method for studying of the drug property differences of TCM prescriptions.
