毛囊干细胞定位、提纯和三维培养的研究进展

摘要 皮肤作为人体最外层的防御系统,具有极强的修复和再生能力。尤其是表皮,由多层角质细胞构成,其基底膜存在着对皮肤再生、损伤修复起决定性作用的干细胞群,称为表皮干细胞(epidermal stem cell,ESC)。它可通过对称分裂或非对称分裂的方式产生定向祖细胞,即短暂增殖(transient amplifying,TA)细胞,继而分化为表皮细胞、毛囊和皮脂腺。皮肤ESC这种特有的生物学特性,使之不仅对皮肤的动态平衡的维持发挥作用,而且对皮肤的损伤修复以及皮肤遗传性疾病、皮肤肿瘤等疾病的基因治疗具有十分重要的意义。
