冈田酸诱导早熟染色体凝聚的辐射剂量效应关系研究 被引量:4

摘要 为研究冈田酸(Okadaic acid)诱导的染色体凝聚与辐射剂量之间的剂量效应关系,采用不同剂量(0、0.5、1.0、2.0、3.0和5.0Gy)的X射线照射离体人外周血,培养48h,终止培养前2h加入Okadaic acid诱导早熟染色体的凝聚,观察分析其诱导的早熟凝聚染色体(Premature condensed chromosome,PCC)的断片率,并与常规染色体畸变分析法进行比较。结果表明,冈田酸诱导的早熟染色体凝聚方法与常规染色体法相比可获得更高的分裂指数,冈田酸法染色体凝聚断片率与外照射剂量之间存在良好的线性关系。 In order to Study the effect-dosage relationship between the PCC induced by Okadaic acid and the IR dosages, human peripheral blood in vitro was irradiated by X-rays at different doses (0. 0.5. 1.0. 2.0. 3.0. 5.0Gy) cultivated for 48 hours and added with Okadaic acid two hours before the end of culture. Chromosome aberrations frequencies was analyzed and compared with that induced by the conventional chromosome assay and their dose-response curves were fitted. The results show that the mitotic index of the PCC induced by Okadaic acid is higher than that of the conventional chromosome assay. And the fragment rate of chromosome condensation induced by Okadalc acid has a favorable linearity relationship with external radiation doses.
