鼻粘膜损伤对风疹病毒再感染的影响

摘要 目的研究鼻粘膜损伤对风疹病毒再感染的影响。方法把风疹病毒IgG抗体阳性的144只豚鼠分为A、B、C、D 4组,A组仅破坏鼻粘膜上纤毛和纤毛细胞及少部分粘液毡,B组则进而破坏1/3的鼻粘膜,c组则进而破坏1/2的鼻粘膜,D组为对照组。再把A、B、C、D组又各分为3亚组,分别用4.5×10^2、4.5×10^3、4.5×10^4 IU/mL的风疹病毒悬液进行鼻腔内感染,7~10天后各组豚鼠断头采血测风疹病毒以及CD4^+、CD8^+细胞以及CD4^+/CD8^+比值,风疹病毒IgM抗体阳性者定量测血清风疹病毒RNA含量;同时取鼻粘膜,观察纤毛及纤毛细胞的情况,以及对鼻粘膜进行免疫组化和HE染色,计数粘膜中CD4^+/CD8^+细胞以及CD4^+/CD8^+比值。结果同种浓度的病毒悬液感染的各组中风疹病毒IgM抗体阳性率以及血液中风疹病毒RNA含量随鼻粘膜损伤程度加大而升高,鼻粘膜损伤各组中血液及鼻粘膜组织中CD4^+/CD8^+细胞以及CD4^+/CD8^+比值明显低于非损伤组,且随鼻粘膜损伤程度加大而降低。结论鼻粘膜损伤易于风疹病毒的生长增殖和再感染发生,从而增大了病毒的再感染率,因此及时治疗修复患者损伤的鼻粘膜可降低风疹病毒的再次感染机会。 Objective: To study the effect of nasal mucosa damage on re -infection of rubella virus. Method: Divide 44 guinea pigs with rubella virus that have IgG positive antibody into four groups (A, B, C and D group). In group A, only destroy the cilia, ciliated cells and a little mucus carpet of the nasal mucosa. In group B destroy 1/3 of the nasal mucosa. In group C destroy 1/2 of the nasal mucosa. And the A, B and C group were divided into 3 smaller groups respectively. We used 4.5×10^2、4.5×10^3、4.5×10^4 IU/mL suspension of rubella virus to cause infection in their nasal. 7 to 10 days later the guinea pigs were sacrificed for blood by which we tested the ratio of rubella virus, CD4^+, CD8^+ cells and CD4^+/CD8^+ ratio, compared with groupt test and chi - square test. Results: The same concentration of virus - infected cells in each group rubella virus IgM antibodies in the blood as well as the positive rate of rubella virus RNA content with the nasal mucosa injury - rise and increase the degree of injury, damage the nasal mueosa blood group and nasal tissur CD4^+, CD8^+ cells and CD4^+/ CD8^+ ratio was signifi- cantly lower than non - injury group, with the nasal mucosa and increase the degree of injury and reduce. Conclusions: Nasal mucosa injury helped the increase of rubella virus and re - infection, which increases the virus infection rate. Therefore, curing the patients with injured nasal mueosa can reduce the chances of rubella virus reinfection.
