NF-κB抑制剂PDTC对CCL19调控人头颈鳞癌细胞增殖活性的作用 被引量:4

摘要 目的探讨核因子κB(NF-κB)抑制剂吡咯烷二硫代氨基甲酸盐(PDTC)在CCL19调控人头颈鳞癌细胞活性中的作用。方法依次使用不同浓度PDTC、CCL19、顺铂处理人头颈鳞癌转移细胞PCI-37B,应用TransAMTM NF-κB p65试剂盒、噻唑蓝比色(MTT)、流式细胞仪等方法观察NF-κB活性变化,及其与细胞生长抑制、细胞周期、凋亡的关系。结果不同浓度PDTC可以使CCL19诱导的NF-κB活性显著降低(P<0.05),同时恢复CCL19作用后的细胞对顺铂化疗的敏感性,使细胞生长抑制率、细胞G1期比例、凋亡率等显著上升(P<0.05)。结论PDTC能阻断CCL19对人头颈鳞癌细胞活性的调控、促使细胞死亡。 Objective To study the role of PDTC in CCL19 regulating the activity of head. and neck squamous cancer cells. Methods The cells were treated with PDTC, CCL19 and cisplatin in order at different concentrations, and then the growth arrest was assayed by MTT, the cell cycle and the apoptosis were determined by flow cytometry (FCM),and the NF-κB activity was observed by TransAMTM NF-κB p65 kit. Results PDTC inhibited NF-κB activity in CCLI9 group in a dose-dependent manner (P〈 0.05),brokdown the growth arrest, and increased apoptosis and G1 phase percentage of cell-cycle which was induced by cisplatin in the cells pretreated by CCL19 (P 〈 0.05). Conclusion PDTC can block the regulation of CCL19 in the activity of human head and neck squamous cancer cells.
