海马内微注射NGF对小鼠学习记忆的影响 被引量:2

INFLUENCE OF NGF BY INTRAHIPPOCAMPAL INJECTION ON LEARNING AND MEMORY IN MICE

摘要 本实验观察了海马内多次注射神经生长因子(NGF)对小鼠学习记忆的影响,包括剂量-效应关系和时间-效应夫系以及3H-Leu掺入突触体蛋白质的示踪研究。结果表明:(l)与注射人工脑脊液(ACSF)的对照组相比,在五种剂量(0.01,0.05,0.l,0.5,1.0pg/pl,双侧海马内注射,每次1.0pl,共注射4次,累积剂量为0.04,0.2,0.4,2.0,4.0pg)中除0.01pg/外,其他4种剂量的NGF均能显著或极显著地提高小鼠Y一迷宫分辨学习的24h记忆保持率,其中0.05pg/pl剂量的效果最佳;(2)最佳剂量的NGF易化记忆的效应在训练后24h最好,96h出现明显的减退(P<0.05);(3)最佳剂量的NGF能使行为训练后小鼠脑突触体蛋白质的3H-Leu掺入量增加。以上结果说明,适当剂量的NGF在一定的时间内具有增强记忆的功效,并能促进脑内突触体蛋白质的合成。 The innuences of NGF by direct intrahippocampal injechon on discriminatioo learning andmemory in Y-maze and the incorporation of 3H--Leu into synaptosomal proteins were observed in young mice.The resultS are as follows (l) Ivhcn NGF of different doses (0.05, 0.l, 0.5, l.0 μg/ μl, the cumulative doses were0.2, 0.4, 2.0, 4.0 μg respectively) were applied, the memory consolidation was improved signincantly as comparedwith the ACSF control (P< 0.01). Among them the 0.05 μg/ al group was most effective. NGF at 0.01 μg/ alhad no significant effect. The faciliating effect of this dose at 24 hours began to decrease significantly (P< 0.05)at 96 hours after training. The NGF at 0.05μg/ al could enhance the incorporation of 3H-Leu intosynaptosomal proteins of the cerebral cortex very significantly (P< 0.01). The results suggest6d that the suitsbledose of NGF could improve the memory consolidation and the syuaptosomal protein synthesis in the brain ofmicee.
