三种常用局部麻醉药对兔离体气管平滑肌收缩作用的影响

摘要 目的观察三种常用局部麻醉药对乙酰胆碱(Ach)诱导的兔离体气管平滑肌收缩作用的影响。方法取兔气管环,纵行切断软骨环,剪成5~8mm×10~15mm条块,浸浴在K-H液中,气管一端连接张力换能器,另一端固定在浴槽的底端。实验一:每个标本用10-3MAch诱导预收缩至张力最大值并记录,然后每隔5min加入不同浓度的局部麻醉药,记录每一浓度5min内的张力最小值。实验二:在K-H液中预先加入不同种类固定浓度的局麻药,浸浴10min,然后每隔5min加入不同浓度的Ach,记录每一浓度5min内张力最大值。结果实验一:3×10-3M丁卡因能明显抑制Ach诱导的收缩;5×10-3M罗哌卡因对预收缩有抑制作用(P<0.01),并且与同浓度的利多卡因比差异有统计学意义(P<0.05)。实验二:利多卡因与罗哌卡因、丁卡因比较,减弱抑制气管平滑肌收缩作用(P<0.05)。随着Ach浓度的增加,丁卡因表现出抑制气管平滑肌收缩作用的持久性,25min时与利多卡因组比差异有统计学意义(P<0.05)。结论三种局部麻醉药对兔离体气管平滑肌的收缩均有一定的抑制作用,但以罗哌卡因最为明显,利多卡因其次。丁卡因作为预防性应用对气管平滑肌的收缩抑制作用较强,并且作用维持较持久。
