大鼠神经病理性疼痛模型血清和脑脊液补体C3的动态变化 被引量:3

摘要 目的观察大鼠神经病理性疼痛(NPP)模型的补体变化特点,探讨补体活化在NPP发病免疫机理中的作用和地位。方法健康雄性SD大鼠20只随机均分为假手术组(S组)和慢性坐骨神经压迫损伤组(CCI组),CCI组大鼠左侧坐骨神经给予松结扎,S组只暴露左侧坐骨神经不予其它处理。用免疫比浊法测定两组大鼠术前和术后第1、2、3、4、5、6、7天血清和脑脊液补体C3的含量。结果两组大鼠血清补体C3含量手术前后差异均无统计学意义。与术前相比,CCI组大鼠脑脊液补体C3含量明显升高(P<0.01),并在建模后第6天达高峰;而S组大鼠脑脊液补体C3含量无明显变化。结论外周神经损伤后,中枢神经系统存在补体异常活化现象,补体的这种变化特点可能在NPP的发生发展中发挥作用。 Objective To oberve the dynamic changes of complement 3 in rats with chronic left sciatic nerve constriction injury(CCI) ,and investigate the effects of abnormal activation of complement protein on the pathological mechanism of neuropathie pain(NPP). Methods Twenty healthy male Sprague Dawley rats were divided randomly into sham-operation group and CCI group. The left sciatic nerve was ligated loosely in rats of CCI group,and that was not in rats of sham-operation group. The levels of complement protein 3 in serum and cerebrospinal fluid (CSF) were determined at pre--operation and 1, 2,3,4,5,6,7 days after operation by immunoturbidimetry. Results The level of complement 3 in serum in both groups was not changed significantly after ligature. Compared with pre-operation,lhe level of complement 3 in CSF in CCI group begin to increase after operation, and reached peak level on the 6th day after operation. However,the level of complement 3 in CSF in sham group exhibited no remarkable change after operation compared to that before. Conclusion The complement components in central nerve system were activated abnormally after the peripheral nerve injury,whicb may play a role in NPP.
