姜黄素对TGF-β1诱导人肾小管上皮细胞增殖的影响 被引量:7

摘要 目的通过观察姜黄素(Curcumin,Cur)对转化生长因子-β1(TGF-β1)诱导人肾小管上皮细胞(HK-2)增殖的影响,探讨姜黄素在防治肾间质纤维化方面的作用机制。方法将HK-2细胞用含10%胎牛血清的DMEM/F12(1∶1)培养基培养;实验分为5组:空白对照组、TGF-β1组(TGF-β110μg/L)、干预1组(TGF-β110μg/L+Cur1μmoL/L)、干预2组(TGF-β110μg/L+Cur5μmoL/L)、干预3组(TGF-β110μg/L+Cur10μmoL/L)。在倒置显微镜下观察各组细胞形态的改变,并通过MTT法检测姜黄素对细胞增殖的影响。结果TGF-β110μg/L能显著诱导人肾小管上皮细胞增殖,与空白对照组相比有显著性差异(P(0.05),并随着处理时间延长而作用增强,但和姜黄素共同作用后,其促细胞增殖作用受到显著抑制(P(0.05)。结论姜黄素能够维持HK-2细胞形态的稳定性,防止细胞纤维样改变,在一定程度上具有抑制TGF-β1诱导细胞增殖的作用,这可能是其防治肾间质纤维化的机制所在。 Objective To observe the influence of curcumin on proliferation of human renal epithelia (HK-2) induced by transforming growth factors-β1 (TGF-β1), and discuss the mechanism of preventing and treating renal fibrosis by curcumin. Methods HK-2 ceils were cultured with culture medium containing 10% of fetal bovine serum {FBS, DMEM-F12 (1 : 1 ) }. The samples were divided into 5 groups, including the blank control group, TGF-β1 group ( 10 μg/L of TGF-β1), intervention group 1 ( 10 μg/L of TGF-β1 + 1 μmoL/L of curcumin), intervention group 2 ( 10 μg/L of TGF-β1 +5 μmoL/L of curcumin ), and intervention group 3 (10 μg/L of TGF-β1 + 10 μmoL/L of cureumin ). The morphological changes of cells in every group were observed with inverted microscope and the influence of curcumin on cell proliferation by using MTT assay. Results The proliferation of HK-2 ceils was induced significantly by 10 μg/L of TGF-β1 with a significant difference compared with the blank control group (P 〈0.05 ). This effect increased with treatment time passing, but when TGF-β1 and curcumin were combined and effected on the cells this effect was inhibited obviously (P 〈 0.05 ). Conclusion Curcumin can maintain the morphological stability of HK-2 cells, prevent ceils from fibrosis, and inhibit cell proliferation induced by TGF-β1 to some extent. It may be the mechanism of preventing and treating renal fibrosis by curcumin.
