有机溶剂联合酶法处理对油菜花粉萌发孔通透性改善的研究 被引量:3

摘要 本实验研究了先用有机溶剂处理,再经酶法处理油菜花粉,改善油菜花粉萌发孔的通透性。结果表明,先经石油醚处理,再用中温淀粉酶和复合纤维素酶处理花粉后,每克花粉上清液中可溶性糖含量最高可达到0.354±0.054g,与空白相比提高了40%;经石油醚处理,再用复合蛋白酶和复合纤维素酶处理花粉后,蛋白溶解指数最高达到0.76±0.04;通过显微镜可以观察到经石油醚处理,再用复合蛋白酶和复合纤维素酶处理后,花粉萌发孔变大,可以非常明显改善花粉萌发孔的通透性。 Rape seed pollen was firstly pretreated with different organic solvents and then treated with different enzymes to improve the permeability of its aperture. The experimental results indicated that after the treatment with medium temperature α - amylase (BAN, Bacterial Amylase Novo) and cellulase (Celluclast, Novozyme), the soluble sugar content in the supematant of 1 g rape seed pollen treated with petroleum ether is 0.354±0.054 g, increasing by 40% compared with the control sample. After the treatment with protease (Protamex) and cellulase (Celluclas0, the protein solution index of rape seed pollen treated with petroleum ether is up to 0.76±0.04, and it was observed that the aperture of the rape seed pollen opens more greatly under microscope. As a conclusion, the aperture of the rape seed pollen is improved obviously.
