摘要
目的观察高盐饮食对不同雄激素状态大鼠血压的影响及与肾素血管紧张素系统(RAS)的关系。方法将雄性 Wistar 大鼠30只随机分为假手术组、去势组、去势后补充睾酮组(补充睾酮组,26.7 mg/kg),每组10只。各组均以8%NaCl 颗粒饲料喂养8周,分别在实验前、实验4周和8周后测尾动脉脉血压,8周后用放免法测外周血清睾酮和血浆肾素活性(PRA)、血管紧张素Ⅱ(AngⅡ)水平。结果 1)在高盐饮食8周后,假手术组和补充睾酮组血压明显升高[假手术组:(137.3±4.0)比实验前:(117.5±5.9)mmHg;补充睾酮组:(134.4±5.2)比实验前:(116.6±7.7)mmHg,P 均<0.01],而去势组血压无明显变化[(11 9.4±8.6)比实验前:(11 5.3±6.6)mmHg,P>0.05]。2)与血压升高相对应,假手术组和补充睾酮组血浆 PRA 及 AngⅡ水平较去势组明显升高[PRA:假手术组(5.90±0.77)比补充睾酮组(5.69±0.47)比去势组(4.90±0.54)mol/(L·h),P 均<0.05;AngⅡ:假手术组(508.1±52.5)比补充睾酮组(493.6±21.1)比去势组(403.6±49.2)ng/L,P 均<0.01)]。结论高盐喂养对雄性大鼠的血压升高作用需要有雄激素的参与,且与 RAS 有关。
Objective To study the effects of high salt intake on blood pressure and renin-angiotensin in male rats with different plasm androgen levels. Methods Thirty male Wistar rats were randomized to sham(n=10) or operated( n=10 ), or castration( n=10 ), or testosterone replacement after castarion ( 26.7 mg/kg, n = 10 ) and fed with 8% NaCl for 8 weeks. Tail arterial pressure were recorded before, 4 and 8 weeks after experiment. Serum PRA, plasma angiotensin Ⅱ (Ang Ⅱ ) and testosterone (T) were determined by radioimmunoassey respectively. Results After 8 weeks high salt dietary, blood pressure was significantly increased in sham and testosterone replacement rats(Sham operation group:137.3±4.0 vs the basal line:117.5 ± 5.9 mmHg, testosterone replacement group: 134.4±5.2 vs the basal line:116.6±7.7 mmHg, P〈0. 01), while little changes in BP was shown in castarted rats ( 119.4 ± 8.6 vs the basal line : 115.3 ± 6.6 mmHg, P〉0.05). Concomitantly, sham operation or testosterone replacement rats had higher PRA and plasm AngⅡ content compared with castrated rats (PRA: Sham operation 5.90±0. 77 vs testosterone replacement group: 5.69±0.47 vs castrated rats: 4.90±0.55 mol/(L· h), P〈0.05; plasma Ang Ⅱ: Sham operation: 508.1±52.5 vs testosterone replacement group: 493.6±21.1 vs castrated rats: 403.6±49.2 ng/L, P〈0.01). Conclusion Androgen seems to involve the mechanism by which the BP elevation induced by high salt diet.
