摘要
目的:了解缺氧复氧对培养新生大鼠心肌细胞超微结构的影响及其黄芪皂甙的保护作用。方法:实验细胞分为三组:对照组(以Hank液代替培养液培养)、损伤组(以氮饱和无氧无糖Hank液密闭培养3h后复氧)及保护组(Hank液中加入25mg·L-1黄芪皂甙,余同损伤组)用电镜观察各组超微结构。结果:电镜下,损伤组心肌细胞的细胞形态、细胞膜、细胞核、线粒体等结构均可发生明显的改变,而黄芪皂甙保护组心肌细胞超微结构的改变则明显较损伤组轻。结论:黄芪皂甙具有减轻实验性缺氧复氧对培养的心肌细胞损伤的作用。
Purpose To study the hypoxia/reoxygenation ultrastructural damage in the cultured new born rat cardiomyocytes and the protective effects of astragalus saponin. Methods The cells were divided into three groups, (1) control, (2) hypoxia/reoxygenation damage and (3) astragalus saponin pretreated. The ultrastructural changes of each group were observed under SEM and TEM. Results The myocardial ultrastructural damage, especially in mitochondria and membrane system, was more significant in the hypoxia/reoxygenation group than in astragalus saponin pretreated group. Conclusions It is suggested that astragalus saponin might decrease the damage of cardiomyocytes undergoing hypoxia/reoxygenation.
