摘要 目的:进一步研究GAP43(生长相关蛋白43)在周围神经损伤和再生中的表达部位及其作用。方法:采用免疫电镜技术对6只大鼠坐骨神经损伤和再生中的GAP43表达情况作初步观察。结果:电镜观察发现大鼠腰骶髓神经元、背根神经节神经元、胶质细胞等胞浆和胞核中均有密集的GAP43免疫反应颗粒沉积。结论:据结果和笔者先前的GAP43免疫组化研究,发现周围神经损伤和再生中,可引起GAP43较广泛的表达;其表达以神经元胞体为主,逐渐向损伤处运送,在神经再生中可能起着重要作用。 Objective:To study the localization of GAP 43 expression and its roles during neuronal regeneration.Methods:Immunoelectron microscopic technigue was usct to expore the location of GAP 43 one week after sciatic nerve transection injury in six rats.Results:Dense immunoreactive particles were found under electron microscope in cytoplasm and nuclei of the cell body of neuron (both the lumbar spinal cord and dorsal root ganglia) and Schwann cell in myeline sheath of the proximal stump of the sciatic nerve.Conclusions:The results in this study and in previous studies demonstrate that GAP 43 can be expressed extensively.It is synthesised mainly in the body of neuron and transported along axon to the site of injury.Although its function has yet to be established,it suggests that GAP 43 plays a critical role in axonal growth and regeneration.
