摘要
采用高效HPLC色谱法测定发酵液中的甜菜碱含量。方法:采用hypersil NH_2化学键合色谱柱(4.6 mm×250mm,5μm),以乙腈:水(65:35)为流动相,示差折光检测器检测,外标法定量。甜菜碱浓度在48.42μg/mL~629.46μg/mL范围内,其峰面积与浓度具有良好的线性关系,相关系数为0.996,平均回收率100.89%。实验证明,此方法适用于发酵液中甜菜碱含量的检测。
The betaine content in fermentation fluid was determined by high performance liquid chromatography. A hyperdil-NH2 chemical-bonded column (4.6mm×250mm,5um) was used with acetonitrile-water (65:35) as mobile phase and a refractive index detector was used for detetion. The external standard method was used for quantification. A good linear relationship was maintained over the range of 48.42-629. 46ug/mL with r=0. 996. The average recovery was 100.89%.
