人血小板生成素对小鼠巨核细胞生长及血小板产生体内外作用研究 被引量:1

Effects of Human Thrombopoietin on Mice Megakaryocytopoiesis and Thrombocytopoiesis in Vivo and in Vitro

摘要 采用血小板计数、巨核细胞形态观察及计数、巨核细胞集落培养技术,观察从人尿中纯化的天然血小板生成素(TPO)对Balb/c,h鼠巨核细胞生成及血小板产生的作用。小鼠随机分四组,Ⅰ组为对照组,Ⅱ、Ⅲ、Ⅳ组为TPO低剂量组(10U/R)中剂量组(50U/只)和高剂量组(100U/只),结果显示TPO能显著提高外周血液中的血小板数(P<0.01)和骨髓中的巨核细胞数(P<0.01),且有巨核细胞的脑体和胞核的显著增大的作用,在体外对巨核细胞集落形成有明显增加作用(P<0.01)。这些结果表明从人尿中纯化的天然TPO具有促进巨核细胞增殖、分化、成熟及产生血小板的作用为TPO进入临床试验提供了依据。 This model was designed to investigate tbe effects of natural purified thrombopoietin (TPO)from human urine on Balb/c mice megakaryocytopoiesis and thrombocytopoiesi s in vivo and in vitro.using blood platelet count, megakaryocyte count, morphology of megakaryocyte and megakaryocyte colonyassay. The results sbowed that TPO significantly increased not only platelet in blood but alsomegakaryocyte in bone marrow (P<0. O1). Megakaryocyte colony assay presented that TPO increased thenumbers of CFU-Meg in vitro. The dose dependent relation with TPO on mice megakaryocytopoiesis andthrombocytopoiesis in vivo and in vitro was confirmed. It was suggested that natural purified TPOmay play an important role in megakaryocytopoiesis and thrombocytopoies is. The clinical applicationof TPO to treat patients with thrombocytopenia will be advanced as the results this study.
