摘要
应用体外集落生成方法,我们对10例再生障碍性贫血(再障)和13例正常对照在体外培养粒-单集落形成单位(CFU-GM)。结果表明:重组人干细胞因子(rhSCF)和重组人粒-单集落刺激因子(rhGM-CSF)均刺激CFU-GM生成,二者联合应用比单独应用rhGM-CSF作用明显增强,但rhSCF单独应用作用很小。
There is no therapeutic method which has effect to patients with Aplastic Anemia,severe infection and hemorrhage are always the causes of death. With the study of model rat,absence/defect of stem cell or stem cell factor (SCF) have therefor been postulated as the cause of Aplastic Anemia. To examine the effect of recombinant human granulocyte/macrophage colony-stimulating factor (rhGM-CSF) and recombinant human stem cell factr (rhSCF) to colony-forming unit-granulocyte/macrophage(CFU-GM)of patients withAplastic Anemia, the clonogenic assay was used. The number of colonies of rhGM-CSF and rhSCF to CFU-GM of 10 patients with Aplastic Anemia and 13 normal contrlos showed: (1)rhGM-CSF and rhSCF can stimulate the growth of CFU-GM both of patients with Aplastic Anemia and normal controls, and the two-factor combination stimulate the greatest number of colonies. (2)rhSCF alone had little effect. When it was combinated with rhGM-CSF, much more effect were achieved. (3)To patients whose marrow were severely affected, rhGM-CSF and rhSCF had little effect.
