摘要 目的通过体外实验初步研究抑癌基因死亡相关蛋白激酶(death associat-ed protein kinase,DAPK)在膀胱癌细胞中的迁移和调亡中的作用研究.方法首先采用免疫荧光染色的方法检测DAPK基因在膀胱癌细胞中的定位,然后用脂质体将全长外源性DAPK基因瞬时转染至不表达该基因的T24细胞中,分别采用划痕实验和流式细胞术检测转染前后膀胱癌细胞的迁移能力和凋亡水平的变化.结果免疫荧光染色发现DAPK基因定位于膀胱癌细胞的胞浆和胞膜.划痕实验提示转染DAPK基因的T24细胞迁移能力下降,流式细胞术检测证实DAPK基因转染后T24细胞凋亡率显著提高.结论作为抑癌基因,DAPK在膀胱癌细胞中具有促进细胞凋亡并抑制细胞迁移的能力. Objective To investigate the biological function of death associated protein kinase gene in bladder cancer cells. Methods Immunofluorescence detected the location of DAPK in ScaBER cells. Full length of DAPK plasmid was transfected to T24 cells without DAPK expression, then wound-healing assays of migration was applied. What is more, flow eytometry was carried out to evaluate the apoptosis rate. Results DAPK located in the cytoplasm and the membrane of ScaBER cell. T24 cells transfected with DAPK revealed a slow migration in the wound-healing assay (P 〈 0.05 ). What is more, the DAPK transfected T24 cells displayed a relatively high apoptosis rate (24.15%) comparing to the non-transfected cells (0.09%). Conclusion DAPK can inhibit the migration ability of T24 cell, and also can promote the apoptosis of bladder cancer cell.
