摘要
目的:探讨大鼠溃疡性结肠炎(UC)模型的血清肿瘤坏死因子α(TNF-α)和白介素10(IL-10)的水平。方法:36只雌性SD大鼠随机分为2组,每组18只。模型组用三硝基苯磺酸(TNBS)的50%乙醇溶液灌肠至结肠内,对照组以50%乙醇溶液灌肠。分别于灌肠后的第1天、第7天、第21天经腹主动脉无菌采血,检测血清TNF-α和IL-10的含量。结果:模型组血清TNF-α53.6±10.5pg/mL和IL-10153.6±12.8pg/L含量于灌肠后第1天均较对照组(分别为28.6±7.8pg/mL和112.7±30.9pg/L)明显升高(P<0.05),而灌肠后第7天及第21天两组血清TNF-α和IL-10含量均无明显差异(P>0.05)。结论:UC早期血清促炎细胞因子和抑炎细胞因子水平均明显升高。
Objective: To explore the serum levels of TNF - α and IL- 10 in rat model of ulcerative colitis.Methods: Thirty-six female SD rats were randomized into two groups (n= 18 each). The mode/group (n= iS) received a colonic enema with trinitrobenzenesulfonic acid in 50% alcohol, whereas the control group (n=18) received the enema with alcohol alone. The serum levels of TNF-α and IL-10 were measured on days 1, 7 and 21 in these rats. Results: Significantly higher levels of serum TNF-α and IL-10 in the model group as compared with the controls were shown on day 1 (53.6±10.5 pg/mL vs 28.6±7.8 pg/mL, 153.6±12.8 pg/L vs 112.7±30.9 pg/L, P〈0.05),but not on days 7 and 21 (P〉 0.05). Conclusion: Both pro-inflammatory and anti-inflammatory eytokines may show elevated levels early in ulcerative colitis.
