摘要
研究木鳖子提取物(ECMS)在Zmu-1:DHP和DHP2个豚鼠品系中,对口蹄疫疫苗的免疫增强作用。将0.1mg ECMS和0.5mL O-AsiaⅠ型双价口蹄疫疫苗混合,免疫2个品系豚鼠,以疫苗组和空白组作对照。用间接血凝法测定O型疫苗持续期的血凝抗体效价;ELISA法测定抗O型疫苗VP1结构蛋白的IgG、IgG1及IgG2;ELISA法测定抗AsiaⅠ型疫苗的IgG、IgG1及IgG2;用流式细胞法和ELIS法,分别测定T淋巴细胞增殖率及淋巴因子。实验结果表明,(1)药物处理组豚鼠B、T淋巴细胞,比疫苗组产生明显的特异性免疫反应(P<0.01或P<0.05)。(2)ECMS能诱导FMD O型疫苗产生较高滴度的IgG及IgG2抗体,但维持时间较短,而诱导AsiaⅠ型疫苗产生的抗体滴度较低,但持续时间较长。(3)药物组能诱导T细胞产生较高滴度的IFN-γ。(4)DHP豚鼠中,ECMS对两个疫苗均有较显著的免疫增强作用,而Zmu-1:DHP豚鼠只对AsiaⅠ免疫增强作用较明显。(5)DHP豚鼠的多数免疫功能大于Zmu-1:DHP豚鼠。实验结果证明:豚鼠是研究ECMS免疫功能较适宜的动物模型;豚鼠品系间的免疫反应存在明显地遗传差异;ECMS对O型苗的免疫增强作用较显著,并主要由IgG2和IFN-γ参与免疫反应。
To evaluate the immune enhancement of ECMS to FMD vaccine, guinea pigs of two different breeds were immunized with O type and AsiaⅠ type of bivalent FMD vaccine with or without ECMS. Serum antibody, T lymphocyte and cytokine IFN- γ, IL-4 were analyzed by IH test, ELISA and flowcytometry. The results showed that the specific immtmoreactions of B and T lymphocytes were significantly enhanced by ECMS (P 〈0.01 or P 〈0.05). High levels of IgG and IgG2 were detected for the O type FMD vaccine, but not the AsiaⅠ type FMD vaccine. Strong immune enhancement effect of ECMS to both vaccines was detected in DHP guinea pig, however in Zmu-1: DHP guinea pig, ECMS only exhibited enhancement to the AsiaⅠ vaccine. These results indicated that the DHP guinea pig is more suitable for investigating the immune function of T and B lymphocytes and biological products.
