重点讨论了酶催化活性测定试剂盒研制中需考虑的两方面的问题:单底物和双底物酶促反应最适底物浓度的理论计算.K_(?)的测定方法及底物选择中的其它注意事项,并以实验加以论证;酶偶联法试剂盒中工具酶最适用量的理论计算。讨论了影响工具酶用量的因素等。
This paper centres on two aspects for thedevelopment of a kit for the measurement of enzy-matic catalytic activity:(1)Theoretical calculationof optimal substrate concentration of one-and two-substrates enzyme-catalyzed reaction.Determina-tion method of Michaelis constant(K_m)andother points important to the selection of sub-strate.(2)Theoretical calculation of optimal quantity of tool enzyme and relationships betweeninfluencing factors on the quantity of toot enzyme.
