禽戊型肝炎血清学调查以及与肝脾肿大综合征关系的研究 被引量:17

Serological Investigation of Avian Hepatitis E Virus Infection and its Relationship with Chicken Hepatitis-splenomegaly Syndrome

摘要 禽戊型肝炎病毒(hepatitis Evirus,HEV)是肝脾肿大(hepatitis-splenomegaly,HS)综合征的主要病原。禽HEV与人和猪HEV均属于肝病毒属。我们的前期研究结果表明基因重组禽HEV的ORF2蛋白含有其独有的抗原表位,同时含有与人、猪HEV共有的抗原表位。本研究用禽HEVORF(Open Reading Frame)2-1蛋白作为包被抗原所建立的检测血清抗体的间接ELISA对从山东、广东、黑龙江采集的914份鸡血清进行了血清学检测,用疑似肝脾肿大综合征鸡肝组织感染SPF鸡群,检测攻毒后血清中可能存在的抗禽HEVORF2抗体。检测结果显示健康鸡群的抗体阳性率平均为28.5%,而患有肝脾肿大综合征的鸡群中的阳性率则为43.3%。用疑似肝脾肿大综合征鸡肝组织感染的SPF鸡群血清抗禽HEVORF2抗体的阳性率则高达62.5%。本研究进一步阐明了禽HEV在我国部分地区的流行状况以及与HS的直接关系。 Avian Hepatitis E virus (HEV) is the major causative agent of hepatitis-splenomegaly (HS) syndrome. Avian HEV is a member of the genus Hepevirus,which also includes human and swine HEVs. Our previous research demonstrated that avian HEV ORF2 antigen contains unique epitopes as well as the ones shared with human and swine HEVs. We report in this study the serological investigation of avian HEV infection by using the indirect ELISA with avian HEV recombinant ORF2-1 as the coating antigen. We tested 914 chicken sera collected from Shandong,Guangdong and Heilongjiang provinces. The relationship between avian HEV and HS syndrome was examined by inoculation of SPF chickens with HS syndrome chicken liver samples. The results demonstrated that the average percentage rate of anti-ORF2 antibody from healthy chickens was 28.5%,while in HS syndrome chickens the average positive rate was 43.3%. The SPF chickens inoculated with liver samples from HS syndrome chickens developed anti-ORF antibodies with the positive rate of up to 62.5%. This research further demonstrated the epidemiology of avian HEV in partial areas of China and provided the evidence of direct relationship between avian HEV and HS syndrome.
