摘要
通过CMC平板初筛、摇瓶发酵复筛,从土样中筛选到1株能够产中性纤维素酶的真菌,经18S rRNA基因序列分析初步确定该菌株为瓶霉菌(Phialophora sp.)。该菌株在适宜的条件下摇瓶培养72h,产生的内切葡聚糖酶活力可达到2.95IU/mL。酶学性质初步研究显示,菌株所产内切葡聚糖酶活力的粗酶液最适pH值为6.0~7.0,且在此pH范围内具较高的稳定性;粗酶液的反应温度以40℃左右为宜,且在50℃以下具有较高的温度稳定性;Mn2+、Fe3+对酶反应有促进作用,Hg2+、Pb2+对酶反应有抑制作用。
A strain of neutral cellulase producing fungi was primarily screened with CMC plating and flask shaking fermentation and isolated from soil. The strain was identified as Phialophora sp. based on the 18S rDNA secluence, analysis. Under optimum conditions, the neutral cellulase activity (CMCase) was 2.95 IU/mL after shaking flask culture for 72 h. The optimal temperature and pH for the enzymatic reaction were 40℃, 6. 0-7.0. Moreover, the results showed that high activity and good stability under 50℃ at pH 6.0-7. 0. The activators of the enzyme were Mn^2+ , Fe^3+ whreas its inhibitors were Hg^2+ , Pb^2+
