摘要 目的:研究基质金属蛋白酶抑制剂GM6001对兔眼滤过性术后抗瘢痕作用。方法:取青紫兰兔24只,双眼行小梁切除术。术后右眼结膜下注射GM6001,左眼注射PBS作为对照,术后测量眼压,观察结膜、角膜、前房、晶状体情况,通过免疫组化方法标记平滑肌肌动蛋白抗体α-SMA阳性细胞数,反映滤过道成纤维细胞增殖情况。结果:术后实验组较对照组眼压低,各时间点实验组α-SMA阳性细胞数均低于对照组(P<0.05),差别有统计学意义。结论:GM 6001具有抑制兔青光眼滤过术后滤过道中成纤维细胞过分增生,减少瘢痕形成的作用,延长了功能性滤过泡存活的时间。 Objective: To study the antiproliferative effects of matrix metalloproteinases inhibitors GM6001 during filtration surgery on rabbit eyes. Methods:Twenty-four pigmented rabbits were employed auto- control. All rabbits(48 eyes)underwent trabeculectomv. The treated eyes were injected subconjunctivally with GM6001 and the control eyes were injected subconjunctivally with PBS after surgery. We observed the intraocular pressure (IOP)and inflammatory restion, α- SMA in filtering pathway were detected by using immunohi stochemical assay. Results: Intraocular pressure( IOP) was significantly lower in treated group (GM6001)than in the control group after surgery. There were statistically Significant differences in the expression level of the α - SMA between two groups(P 〈 0.05 ). Conclusion: GM6001 have the efficacy of inhibit the fibroblast cells proliferation in filtering pathway, show better IOP control and bleb survival in this experimental model of filtering surgery.
