摘要
目的:探讨IL-12在约氏疟原虫感染中的作用。方法:尾静脉注射法感染IL-12基因敲除小鼠和B6小鼠约氏疟原虫(17XNL),于感染后的第0、2或4、6天取小鼠尾静脉血,检测血清中的细胞因子;从感染后的第2天起,隔日检测小鼠虫体血症水平。结果:与B6小鼠相比,IL-12基因敲除小鼠在感染后虫体血症水平增长迅速且水平高;清除感染需时较长,但仍能完全清除虫体而存活。在感染后的第0、2、6天血清中无IL-12、IFN-γ,而B6小鼠IL-12I、FN-γ(D4)水平均较高。两者在TNF-α水平上无差别。结论:尽管IL-12对小鼠感染约氏疟原虫后的存活很重要,但无IL-12的小鼠感染约氏疟原虫后仍能存活。
Objective: To investigale the influence of IL - 12 on Plasmodium yoelii infection. Methods: IL - 12 kockout (KO) mice and B6 mice were infected intravenously with Plasmodium yoelii 17XNL parasitized red blood cells, their parasitemia were measured every two days from the second day after infection respectively. Cytokines in the serum of these mice were measured on the day 0, 2 or 4 and 6 after infection. Results:The parasitemia of IL - 12 kockout mice went up faster and higher, it took more time to clean the parasite, but cleared completely and could survival compared with B6 mice. There are lots of IL- 12 and IFN-γ on day 4 after infection in B6 mice with 17XNL, whereas, no in IL- 12 KO mice. There is no difference between the TNF-α. Conclusion: Even though IL- 12 is very important for survival, the mice would survival from the Plasmodium yoelii 17XNL infection without IL- 12.
