摘要
在逆转录病毒介导下将TNF基因转染入小鼠LAK细胞中,采用G418抗性筛选和有限稀释法,将LAK细胞克隆化,并筛选出3株分泌不同水平TNF的LAK细胞克隆。结果表明,这3株LAK细胞克隆分泌的TNF水平、增殖能力和杀伤活性均显著高于对照组LAK细胞。而高分泌株的LAK细胞克隆,其增殖能力和杀伤活性最高;中分泌株次之;低分泌株相对较低。抗TNF单抗可显著抑制此3株LAK细胞体外杀伤活性,表明其杀伤活性升高是由基因转染后所表达的TNF介导的,所表达TNF水平越高,其增殖能力和杀伤活性越高。
TNF gene was transfected into murine LAK cells by retrovirus.Three groups of LAK cells were cloned by G418 resistant selection and limiting dilution techniques.The results showed that TNF genetransfected LAK cell clones secreted higher levels of TNF than LAK cell clone in the control group.Proliferation and cytotoxicity were found to be greatest in the TNF genetransfected LAK cells clones with the highest secreting TNF level and the smallest with the lowest secreting TNF level.The cytotoxicity of TNF genetransfected LAK cell clones was markedly inhibited by antiTNF monoclonal antibody,indicating that the above augmentation was mediated by TNF secreted by transfected LAK cell clones.The higher secreting TNF level corresponds with proliferation and cytotoxicity.
