摘要
目的研究骨髓间充质干细胞(MSCs)对异种混合T淋巴细胞增殖的影响及其机制。方法采用密度梯度离心法分离豚鼠MSCs;免疫组织化学法检测MSCs膜表面CIN5和CD44的表达;流式细胞术检测MSKCs周期;CCK-8法检测和绘制MSCs的生长曲线;观察MSCs和其培养上清对体外异种淋巴细胞混合培养的影响。结果MSCs膜表面CIN5表达阴性、CIN4表达阳性;分析MSCs周期显示,MSCs中20%左有处于S期、G2期和M期,其余80%左右均处于G0期和G1期。MSCs和其培养上清均能明显抑制异种T淋巴细胞混合培养的增殖反应(P〈0.05),加入白细胞介素2(IL2)能逆转MSCs的抑制作用(P〈0.05)。结论MSCs的鉴定可根据细胞形态,膜表面标志物来确定。MSCs及其培养上清对异种淋巴细胞混合培养的增殖反应有显著的抑制作用。
Objective To observe the influence of MSCs on heterologous I cell prolllerauon ano the mechanism. Methyls The MSCs obtained by centrifugation over Percoll was cultured in vitro. Cell surface epitopes of CD45 and CD44 were analyzed by immunohistochemistry. The cell cycle of MSCs was analyzed by flow cytometry. Cell growth curve was analyzed by CCK 8. Effects of MSCs or the culture supernatant on heterologous T celt proliferation were detected by CCK-8. Results lmmunohistochemistry assay revealed MSCs were positive for CD44. but negative for CD45. 20% of MSCs were in S, G2 and M phases and 80 74 in G0 and G1 phases. MSCs and the culture supernatant could significantly inhibit heterologous T cell proliferation (P〈0.05). Addition of IL-2 could reverse the in hibitory effect by MSCs (P〈0.05). Conclusion MSCs can be identified by virtue of cell morphology, membrane phenotype and MSCs may play a role in significant immunosuppression of heterologous T cell proliferation.
