重组毕赤酵母SMD1168/pGAPZaA-VAP1稳定性及发酵表达WSSV-VAP1 被引量:1

摘要 研究发酵时间和接种量对WSSV-VAP1重组菌蛋白表达量的影响及重组毕赤酵母中外源基因的稳定性。结果表明,构建的重组菌SMD1168/pGAPZaA-VAP1在发酵72h后,蛋白表达量占总蛋白10.45%,控制接种量对基因工程菌Pichia pastoris表达WSSV-VAP1非常重要。外源基因的插入对宿主菌的生长未产生明显的影响,重组质粒在毕赤酵母SMD1168中很稳定。 The effect of inoculation and fermention time on WSSV-VAP1 expression in recombinant Pichia pastoris was studied. The result showed that the percentage of expression protein was 10. 45% of total supernant protein. The volume of inoculation was an important factor for WSSV-VAP1 expression. Stability of WSSV-VAP1 gene in recombinant P. pastoris showed that the transformation of WSSV-VAP1 gene had little effect on the growth of the host cells. The recombinant plasmid had excellent stability in P. pastoris SMDl168.
