摘要
目的:观察人肺癌裸鼠移植瘤整合蛋白表达与其转移的相关性。方法:以ABC免疫组化染色,对转移力由低到高依次为PLA-801C、Anip973、PLA-801D、PLA-801DL四株人肺癌裸鼠移植瘤作整合蛋白亚单位表达的观察,并在裸鼠体内观察整合蛋白β1抗体及其配体RGD多肽对移植瘤生长及转移的影响。结果:四株移植瘤均呈α2、α5、α6B阴性。β1的表达除Anip973呈强阳性、PLA-801DL呈弱阳性外,也均呈阴性。但四株移植瘤除PLA-801C外,其余3株均呈α6A强阳性。β1抗体及RGD多肽对PLA-801DL移植瘤的转移不见明显影响。结论:4株人肺癌移植瘤总体上呈β1亚族整合蛋白低表达,表达水平与其转移无显著相关。
Objective: To investigate the relationship between the expression of integrin of transplanted human lung carcinoma and its metastasis in nude mice. Methods: The expression of integrin subunits were observed with ABC immunohistochemistry in four strains of xenografts of human lung carcinoma. The metastatic potential of the four xenografts was from low to high among PLA801C, Anip973, PLA801D and PLA801DL in nude mice. The effects of β1 antibody and RGD peptide on the PLA801DL were also studied. Results: α2,α5 and α6 B were all negatively expressed. The expression of integrin subunit β1 was only strongly positive in Anip973 and weakly positive in PLA801DL. But high expression of α6 B was found in PLA801D, PLA801DL and Anip973 except PLA801C. β1 subunit antibody and RGD peptide exerted no marked effects on metastasis of PLA801DL xenograft. Conclusion: Low expression of integrin β1 subfamily exists in four strains of xenografts of human lung carcinoma but is not correlated to their metastatic capacity.
