黑脊倒刺鲃染色体和同工酶分析 被引量:5

Analysis of Chromosome and Isozymes for Spinibarbus caldwelli

摘要 采用肾细胞短期培养和空气干燥法制备黑脊倒刺鲃(Spin ibarbus caldwelli)有丝分裂中期染色体,并对核型进行分析.结果表明黑脊倒刺鲃的染色体二倍数2n=100,核型公式2n=18m+32 sm+30 st+20 t,臂数NF=150.同时,采用聚丙烯酰胺垂直凝胶电泳技术,对黑脊倒刺鲃的血清、肝脏、心脏、肾脏和肌肉等5种组织的乳酸脱氢酶(LDH)、酯酶(EST)和苹果酸脱氢酶(MDH)的同工酶进行了分析.结果显示,LDH在肌肉组织中高水平表达A基因座位,在其他组织中均表达A和B两个基因座位;EST在肌肉组织中没有表达,在其他组织中有表达,但表现出组织内及组织间广泛多态;而MDH在各种组织中均不表达或弱表达.这表明黑脊倒刺鲃的这几种酶存在组织特异性. The chromosomes and karyotype of the Spinibarbus caldwelli were analyzed by short-term in vitro culture of kidney cell and air-drying technique. The results showed that the chromosome number in diploid was 100 (2n = 100), the karyotype formula 2n = 18m +32sm +30st +20t, the fundamental arm number NF = 150. In the same time, the three isozymes, Lactate Dehydrogenase (LDH), Esterase (EST) and Malate Dehydrogenase (MDH), from 5 tissues (serum, liver, heart, kidney and muscle) of S. caldwelli were analyzed by vertical polyacrylamide gel electrophoresis technique. The results showed that as for LDH, while 2 genes (Ldh - A, Ldh - B) expressed in other tissues, only one gene (Ldh - A) expressed highly in muscle. The EST expressed in all tissues except the muscle, and displayed extensive polymorphism among and within tissues. The MDH didn't express ( or expressed weakly) in the tissues. It suggested the 3 isozymes had different expression patterns in various tissues of S. caldwelli.
