摘要
目的研究上皮-间质细胞转化相关上皮细胞标志因子在C57小鼠Lewis肺癌中的表达以及肺岩宁方对其mRNA表达的影响。方法采用实时PCR观察C57小鼠Lewis肺癌右腋下移植瘤和远处转移灶发生的肺组织中上皮型钙黏附蛋白(E-cad-herin)、α-连环蛋白(α-catenin)、β-连环蛋白(β-catenin)mRNA表达及肺岩宁方对其表达的影响。结果肺岩宁治疗组较模型组肺转移率明显降低(P<0.01);对于各组移植瘤组织中,肺岩宁治疗组及化疗阳性对照组的移植瘤组织中E-cadherin、α-catenin及β-catenin均较模型组升高(P<0.01);模型组肺组织中E-cadherin、α-catenin、β-catenin表达明显低于正常肺组织中的表达(P<0.05),而经过肺岩宁方治疗后,三个标志因子均有显著升高(P<0.05或P<0.01)。结论肺岩宁方具有上调上皮细胞标志因子的作用,其可能通过抑制上皮-间质细胞转化过程的发生而达到抗肿瘤转移的作用。
Objective To study the expressions of epithelial symbol factors including E-cadherin. α-catenin, and β-catenin related to epithelial-mesenchymal transition (EMT) in C57 mice Lewis lung cancer and its mRNA expression affected by Feiyanning Decoction (Decoction for lung cancer). Methods The Real-Time PCR method was adopted to observe the E-cadherin, α-catenin. β-catenin mRNA expression of C57 mice with transplanted tumor in the right armpit and distal metastases and the affection of Feiyanning Decoction on the expression. Results The lung transplanted rate in the Feiyanning group was clearly Iower than that in the model group (P〈0.01). In the transplanted tumor tissues of all groups, the expression of E-cadherin. α-carenin and β-catenin of Feiyanning group and chemotherapy positive control group was all higher than that of model group (P〈0.01). As in the lung tissues of all groups, the expression of E-cadherin. α-catenin and β-catenin of model group was clearly lower than that of normal lung tissues (P〈0.05), but with treatment of Feiyanning Decoction. these 3 symbol factors were all clearly increased (P〈0. 05 or P〈0.01). Conclusion Feiyanning Decoction functions to up-regulate the epithelial symbol factors, playing a role in anti-metastasis of tumors maybe through inhibiting the occurrence of EMT.
