摘要
用离体器官实验法,在离体兔主动脉环和离体豚鼠右心房上研究黄芪甲苷(ASI)对心血管的作用,并与维拉帕米(Ver)的作用相比较。结果:ASI对KCl和CaCl2所致兔主动脉环收缩,呈非竞争性拮抗,PD2值分别为3.11和3.57,ASI的作用弱于Ver。ASI对豚鼠离体右心房自发性节律有抑制作用,此作用能被Ca2+对抗。表明ASI对KCl和CaCl2所致的离体兔主动脉环收缩和对离体豚鼠右心房自发性节律有抑制作用,此作用似乎与拮抗Ca2+有关。
The isolated aortic rings of rabbit and isolated right atria of guinea pig were used for the investigating of the effects of ASI on cardiovascular system. The effects of ASI were compared with those of verapamil(Ver).The results showed that ASI shifted the dose response curve of CaCl 2 and KCl to the right in a nonparallel fashion and decresed their maximal response, showing noncompetitive antagonism.The values of pD 2 were 3.11 and 3.57 respectively.The action of ASI was less in potency than that of Ver.The result also showed that ASI inhibited the normal automaticity of the isolated right atria of guinea pig. This effect can be antagonized by 5 mmol/L CaCl 2. The above information suggests that ASI could inhibite the normal automaticity of the isolated right atria of guinea pig and the CaCl 2 or KCl-induced contraction in the isolated aortic rings of rabbit. The inhibitory effect of ASI seems to be associated with the antagonization of calcium ion.
