摘要 [目的]预测尼罗罗非鱼DMRT蛋白B细胞表位。[方法]以尼罗罗非鱼Dmrt基因序列为基础,按Garnier-Robson法,Chou-Fasman法和Karplus-Schulz法预测其编码蛋白的二级结构;用Kyte-Doolittle法,Emini法及Jameso-Wolf法分别预测B细胞表位。[结果]Garnier-Robson法和Chou-Fasman法的预测结果均表明,在尼罗罗非鱼DMRT蛋白分子N端第31-56、68-75、110-116、209-211区段和第239-243区段可能是α螺旋中心;蛋白N端第95-99、177-183、225-234区段和第251-254区段可能是β折叠中心;按Kyte-Doolittle的亲水性方案、Emini方案和Jameson-Wolf抗原指数方案,辅以对DMRT蛋白的二级结构中的柔性区域的分析,预测DMRT蛋白的B细胞表位,DMRT蛋白N端第13-16、35-38、47-54,84-93、101-109、127-156、166-177区段和第198-201区段附近很可能为B细胞表位优势区域。[结论]该研究结果为DMRT蛋白克隆抗体的制备及探索尼罗罗非鱼性别调控机理提供了参考资料。 [Objective] The research aimed to predict the B cell epitope of DMRT protein in Oreochromis niloticus.[Method] The secondary structure of amino acid sequence of DMRT protein was revealed by Garnier-Robson,Chou-Fasman and KarplusSchulz methods.The hydrophilicity plot,surface probability and antigenic index were obtained by Kyte-Doolittle,Emini and Jameson-Wolf methods,respectively.Based on the above results,the B cell epitopes for DMRT were predicted.[Result] Both the prediction results from Garnier-Robson,Chou-Fasman methods indicate that the α-helix centers of DMRT protein in Oreochromis niloticus are in the N terminal No.31-56,68-75,110-116,209-211 and 239-243;the β-sheet centers of DMRT protein in Oreochromis niloticus are in the N-terminal No.95-99,177-183,225-234 and 251-254.With the assistant of Kyte-Doolittle,Emini and JamesonWolf methods,the B cell epitopes for DMRT were located in or nearby the N-terminal No.13-16、35-38、47-54,84-93、101-109、127-156、166-177 and 198-201.[Conclusion] These results are helpful for preparing antibody of DMRT protein and revealing the sex determination mechanism of Oreochromis niloticus.
