摘要
目的研究冠心Ⅱ号对大鼠实验性心肌梗死的治疗作用,观察心肌梗死发生后血清血管内皮生长因子(VEGF)、碱性成纤维生长因子(bFGF)与B型钠尿肽(BNP)的相关性。方法健康雄性大鼠36只随机选取9只作为假手术组,另27只行冠脉结扎术制造心肌梗死模型,后分为模型组冠心Ⅱ号低剂量组(冠心Ⅱ号3g/kg)冠心Ⅱ号高剂量组(冠心Ⅱ号9g/kg)各9只,术后15日进行心肌梗死面积定量组织学测定,酶联免疫吸附法测定血清VEGF、bFGF和BNP等指标,BivariateCorrelation法分析心肌梗死发生后血清VEGF、bFGF与BNP的相关性。结果给药15日后,冠心Ⅱ号高剂量组心肌梗死范围减小,血清BNP降低,VEGF升高,与模型组比较差异有统计学意义(P<0.01或P<0.05)。结论冠心Ⅱ号可以减轻心肌坏死,可通过调节VEGF、bFGF对心肌梗死发挥治疗作用,心肌梗死后VEGF、bFGF与BNP具有相关性。
Objective To study the therapeutic effect of Guanxin Ⅱ (Prescription for coronary heart disease) on the experimental myocardial infarction of rats and to observe the correlation among the vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF) and B natriuretic peptide (BNP) after the occurrence of myocardial infarction. Methods Of the 36 healthy male rats, 9 were put into the sham operation group. Of the 27 male rats with coronary artery ligation to establish the myocardial infarction models, 9 were put into the model group, 9 put into the Guanxin Ⅱ low dosage group (3g/kg), and 9 put into the Guanxin high dosage group (9g/kg). Fifteen days after the operation, N-BT staining was adopted to determine the area of myocardial infarction, enzyme-linked immunosorbent assay (ELISA) used to determine VEGF, bFGF and BNP, and Bivariate Correlation method was used to analyze the correlation of VEGF, bFGF and BNP. Results With 15 days' medication, the area of myocardial infarction in the Guanxin Ⅱ high dosage group was reduced, BNP decreased and VEGF increased, showing a statistical significance in eomparing with the model group (P〈0.01 or P〈0.05). Conclusion Guanxin Ⅱ can reduce the area of myocardial infarction. It treats myocardial infarction through regulating VEGF and bFGF. And VEGF, bFGF and BNP are correlated with each other after occurrence of myocardial infarction.
