摘要 [目的]探讨用氨基酸自动分析仪建立一种测定烟草中游离氨基酸的方法。[方法]用氨基酸分析仪法测定烟叶中的游离氨基酸并对这些游离氨基酸分别定性定量检测。[结果]氨基酸分析仪法能够分离检测烟叶中的23种游离氨基酸(除了蛋氨酸),其回收率均在78.86%-101.46%,标准偏差均在8%以内,具有较好的重现性和回收率。最优的氨基酸提取方法是:样品过60目筛,用0.005mol/L的盐酸超声萃取30min。对不同的烟叶样品进行检测,发现不同种类烟叶所含有的氨基酸具有一定的规律性。烤烟和香料烟中所含的游离氨基酸以Pro为主,而白肋烟以Asn为主;只有白肋烟中含有氨基酸Tau,而烤烟和香料烟里面几乎检测不到。[结论]氨基酸分析仪法是测定烟叶中游离氨基酸较好的方法。 [ Objective ] A method established for the detection of free amino acids in tobacco using amino acid analyzer was discussed. [ Method ] The free amino acids in tobacco were detected using the amino acid analyzer and these free amino acids were detected qualitatively and quantitatively resp. [ Result] 23 kinds of free amino acids except methionine in tobacco leaves were separated and detected by the amino acid analyzer method, the reproducibility were between 78.86% and 101.46% , and the standard deviation values were below 8%. The method had better reproducibility and recovery. The optimum extraction method of amino acid was as follows : the samples were sieved by 60 meshes treatment and treated by ultrasonic extraction with 0.005 mol/L hydrochloride for 30 min. The contents of amino acids in different varieties of tobacco leaves showed some law after the detecting tobacco samples. The free amino acid contained in flue-cured tobacco and oriental tobacco was mainly praline, while that in burley tobacco was mainly asparagine. The taurine was only contained in burley tobacco and it was nearly not detected in flue-cured tobacco and oriental tobacco. [ Conclusion] The amino acid analyzer method was the better method for detecting the free amino acids in tobacco leaves.
