补肾填精方对肾虚雄性小鼠及其雄性仔鼠睾丸组织形态学的影响 被引量:3

Effect of Bushen Tianjing(补肾填精) Recipe of Histologic Morphology of Tests of Mice of Insufficient Shen Jing(肾经) and their Male Offspring

摘要 目的:观察补肾填精方对肾虚雄性小鼠及其雄性仔鼠睾丸组织学的影响。方法:将30只雄性昆明小鼠随机分为正常对照组、模型组和补肾组。采用"房劳加惊恐伤肾法"造模,从造模次日起,所有小鼠与正常发情雌鼠配对饲养5d,处死,取睾丸。另取出雌鼠待其分娩。仔鼠饲养至6周龄时每组随机取出10只,取睾丸。分别观察父代小鼠及其仔鼠睾丸组织病理学改变。结果:模型组小鼠生精小管边界扭曲,界膜轻度增生。Johnsen积分、生精小管直径及生精上皮厚度都明显低于正常组。其仔鼠睾丸组织呈现出类似改变,只是病变程度略轻。补肾组小鼠及其仔鼠Johnsen积分和生精小管直径与正常组相比均无显著差异。结论:"惊恐、房劳"肾虚模型小鼠受损的生精功能可以传递至没有受到造模因素影响的雄性仔鼠。 Objective To study the effect on the pathohistology of testes of mice of insufficient ShenJing(肾经) and their mal offspring. Methods KunMing mice, 6 weeks old, 30 male and 330 females, were allocated to 3 groups(blank control group, model group, BuShen(补肾) group) randomly. The mice of the model group and the BuShen group were processed by the "fearness combined with excessive sexual hehavior" to made the model of insufficient ShenJing. The mice in BuShen group were lavaged of concentrated solution of BuShenFang, the dosage was 0.16 ml/10g based on their weight. Other groups were treated with physiological saline at the same dosage. This phase would continued 21 days. All the 3 groups were matched - pairs with healthy female mice for 5 days after model making. The microstructure and ultrastructure of testiculus of male mice and their male offspring was observed. Results Compared with normal(9.6 ± 0.46) group, the average Johsen scores of the model group(8.97± 0.65) were decreased significantly( P 〈 0.05) ; the diameter length( 115.27±15.39 μm) and the thickness(26.13 ±5.62 μm) of seminiferous tubule of the model group were decreased too (normal group 127.61 ± 19.16 μm, 33.70 ± 6.82 μm, BuShen group 125.94 ± 17.56μm, 29.5 ± 4.97μm, P 〈 0.05). There was no significant difference between normal group and BuShen group ( P 〈0. 05). The average Johnsen scores (8.88 ± 0.6), the diameter length (116.49 ± 15.18μm) and the thickness (25.86 ± 5.35μm) of seminiferous tubule of F1 of model mice were lower than those of the normal (9.71 ± 0.51, 128.15 ± 18.35 μm, 33.57 ± 6.94 μm). But there was no significant difference of these data between the F1 male mice of model group and BuShen group. Conclusion Fearness combined with excessive sexual behavior can damage the microstructure and ultrastructure of testicutus of model mice, and these pathologic changes can be transferred to their male offspring. BuShen can interrupt this effect.
