摘要
在鸡土从菌(Termitomycesalbuminosus)菌丝体液体深层培养成功的基础上,为进一步对其染色体进行分析,采用液氮研磨、SDS和蛋白酶K消化的方法,从液体培养的鸡土从菌菌丝体中提取出高分子量的染色体DNA,并用紫外吸收光谱、凝胶电泳和限制酶切等方法进行了鉴定。通过Tm值推算出鸡土从菌染色体DNA的G+C含量为43.7%
Chromosomal DNA was extracted and isolated from mycelia Termitomyces albuminosus submerged culture,ground with liquid nitrogen,and treated with SDS and proteinase K.The properties of the DNA were identified by U.V. absorption rate,agarose gel electrophoresis and restriction endonuclease digestion.The Tm of chromosomal DNA from Termitomyces albuminosus determined was 87.2℃.The G+C content was estimated to be 43.7%.
