摘要
应用卵蛋白致敏和刺激小鼠复制过敏性哮喘模型观察血栓素A2合成酶选择性抑制剂OKY-046对抗原引起呼吸道嗜酸性粒细胞(EOS)浸润的影响。结果表明正常对照小鼠支气管肺泡灌洗液(BALF)中未见到EOS;致敏小鼠给予抗原反复刺激后,BALF中EOS急剧增多。OKY-046的不同剂量分别导致BALF中EOS数下降36.9%,58.0%及68.9%。还发现OKY-046抑制EOS对气道的侵润伴随着IL-5和IL-2水平的明显降低,这些结果提示OKY-046通过抑制CD4T淋巴细胞分泌细胞因子从而抑制了EOS在气道的积聚。
An antigen induced airway inflammation mouse model was used to investigate the effects of thromboxane synthetase inhibitor OKY 046 on eosinophil (EOS) infiltration of the respiratory tract Results:no EOS could be found in the bronchoalveolar lavage fluid (BALF) obtained from the normal control mice.However,chronic ovalbumin challenge in sensitized mice induced significant increase of EOS in BALF.Different doses of OKY 046 produced respectively 36 9%,58 0%,and 68 9% decrease in the number of EOS in BALF.Also,the inhibition of EOS infiltration of the respiratory tract was acconpanied by a decrease in both interleukin IL-5 and IL-2 levels These results indicate that OKY 046 prevente airway eosinophil infiltration by inhibiting the secretion of cytokines by CD 4 + T lymphocytes.
