摘要
目的:研究左归丸对骨髓源成体干细胞(adult stem cells,ASC)多向分化的调控作用。方法:以贴壁筛选法分离骨髓源ASC,采用细胞化学和计数等形态学方法,观察骨髓源ASC在体外扩增及多向分化潜能。结果:左归丸Ⅰ号(全方)和左归丸Ⅱ号(全方减去龟鹿二胶)均能诱导骨髓源ASC向成骨细胞、软骨细胞、神经元细胞和神经胶质细胞方向分化。结论:诱导骨髓源ASC向神经元样细胞及神经胶质样细胞方向分化,左归丸Ⅰ号明显地优于左归丸Ⅱ号;但左归丸Ⅱ号诱导骨髓源ASC向成骨样细胞及软骨样细胞方向分化的作用又显著优于左归丸Ⅰ号。
Objective: To investigate the controlled effect of Zuo Gui Wan's compatibility on multi - directional differentiation of the bone marrow - derived adult stem cells (BMDASCs) . Methods: Adherence Screening Method was used to isolate the rat BMDASCs, amplification and multi - directional differentiation's potentiality of BMDASCs in vitro were observed by morphology way, such as cytoehemistry and cell counting. Result: To Zuo Gui Wan I (complete formula) and Zuo Gui Wan II (modificated formula that subtract Gui Jiao and Lu Jiao), both of them can induce differentiation of BMDASCs into osteoblast, cartilage cell, neuron and neurogli- al cell. Conclusion: To induce differentiation of BMDASCs into neuron and neuroglial cell, Zuo Gui Wan I is better than Zuo Gui Wan II, but Zuo Gui Wan 11 excel to Zuo Gui Wan I significantly when induce differentiation into osteoblast, cartilage cell.
