摘要
以聚乙二醇二氨为偶联剂,通过叶酸活性酯和聚合物端基活性酯与聚乙二醇二氨反应,制得叶酸修饰的大分子,再通过乳化法合成载紫杉醇纳米粒,制备叶酸修饰的乳酸-羟基乙酸共聚物纳米粒子。紫外光谱、红外光谱、核磁光谱显示叶酸成功连接在高聚物分子上。所制得的纳米粒粒径(276±12)nm,扫描电镜观察其形态为规整的球形。该方法可成功制备叶酸修饰的乳酸-羟基乙酸共聚物载药纳米粒。
Folate-mediated poly (lactic-co-glycolic) acid was prepared by the reaction of the activated folate ester and activated polymer with the amine group on the poly (ethylene glycol)-bis-amine. Folate-mediated poly (lactic-co-glycolic) acid nanoparticles were then formulated with the emulsion-solvent evaporation method. Results of ultraviolet spectra (UV) spectrum test, infrared (IR) spectrum test and nuclear magnetic resonance (H1NMR) test that the folate was conjugated successfully with polymer. The average diameter of nanoparticles was (276_+12) nm and the Folate-mediated poly (lactic-co-glycolic) acid nanoparticles were spherical in shape as shown under the scanning election microscope. Folate-mediated poly (lactic-co-glycolic) acid nanoparticles for loading paclitaxel could be prepared successfully with the method.
