Establishment of mouse obesity asthma model

摘要 目的探索在高脂饲料诱导肥胖基础上建立哮喘动物模型,并对模型进行评估。方法40只清洁级雌性3周断乳C57BL/6J小鼠按随机数字表法分为4组,每组10只。分别进行普通饲料喂养+NS致敏激发(A组);普通饲料喂养+OVA致敏激发(B组);高脂饲料喂养+NS致敏激发(C组);高脂饲料喂养+OVA致敏激发(D组),比较各组肥胖和哮喘评价指标。结果①C、D组体质量、体长、Lee’s指数、肝脏质量、肝脏/体质量均明显大于A、B组(P<0.05)。②A、C组小鼠肺组织无炎症改变,B、D组炎症浸润明显,其中D组更甚。③支气管肺泡灌洗液细胞总数、嗜酸性粒细胞比例D组明显高于各组,B组明显高于A组(P<0.05)。④B、D组支气管肺泡灌洗液中IL-4分别较A、C组明显升高(P<0.05),IFN-γ则明显降低(P<0.05),D组IL-4/IFN-γ明显高于B组(P<0.05)。结论用不含致敏原的自制高脂饲料喂养,采用OVA致敏、激发C57BL/6J小鼠成功建立了肥胖哮喘模型。 Objective To establish an obesity animal model in mice and evaluate the model. Methods A total of 40 clean 3-week-old weaning female C57BL/6J mice were randomly and equally divided into 4 groups. Group A were fed with normal diet food and sensitized with normal saline ( NS), group B were fed with normal diet and challenged with ovalbumin (OVA). Groups C and D were fed with high fat diet and only the later received OVA challenging. Body weight, liver weight, index of liver/body weight, Lee' s index, lung morphology, cell number of bronchoalveolar lavage fluid, concentration of IL-4 and IFN-γ were measured or calculated. Results ( 1 ) The body weight, liver weight, index of liver/body weight, Lee' s index of group C and D were significantly higher than those of group A and B. (2) Local inflammation was significant in groups B and D, with the later more serious. (3) Total cell count and ratio of eosinophil of bronehoalveolar lavage fluid were highest in group D followed by group B and group A. (4) Groups B and D had a higher concentration of IL-4 and lower level of IFN-γ in comparison with group A and C, and the ratio of IL4/IFN-γ was higher in group D than group B. (5) Two mice respectively from groups C and D had not meet the obesity criterion before challenge, and the other 2 mice from group B and D respectively died during challenge, so they were excluded in this study. Conclusion Mouse obesity asthma animal model can be established in C57BI./6J mice by feeding with self-made high fat food and sensitized and provocated by OVA.
