摘要
为深入了解血液常温保存中红细胞的损伤性变化,采用电子自旋共振(ESR)技术对常温(35℃)保存的人血样品,进行了血红蛋白氧化变性研究。结果显示:35℃保存的全血在保存36小时左右出现高自旋的高铁血红蛋白吸收峰(g=6),且随保存时间的增加而增大。在4℃保存的全血,连续60小时的保存实验均未发现有任何ESR波谱吸收峰出现。35℃保存人血红细胞中,超氧化物歧化酶的活性随保存时间的延长而逐步降低。血红蛋白的氧化变化,可能是由于红细胞内抗氧化系统在血液保存期间抗氧化效力逐步降低造成的。
Studying the denaturation of oxidative hemoglobin of blood preserved at room temperature,it is helpful to overall understand erythrocyte injurious during blood preserved at room temperature.Using electron spin resonance technique,we have researched the denaturation of oxidative hemoglobin of blood preserved at 35℃. We discovered that blood preserved at 35℃, appears ESR absorption of high spin methemoglobin(g=6) after preservation time is 36 hours.The high spin methemoglobin ESR absorption will increase when preservation time increases.When blood is preserved at 4℃,it dose not emerge ESR absorption during 60 hours preservation. SOD activity will gradually decline in erythrocyte of blood preserved at 35℃ when preservation time increases. Oxidative denaturation of hemoglobin is probably caused with gradual declining of antioxidation system effect in erythrocyte.
