摘要
目的研究成年大鼠脑出血模型中发生的神经细胞再生现象及其特点。方法应用雄性Sprague—Dawley大鼠制作脑出血模型,采用蛋白印迹分析、免疫组化染色等方法对5-溴脱氧尿嘧啶核苷(Bromodeoxyuridine,Brdu)及未成熟神经元标记doublecortin(DCX)等进行分析。结果DCX的表达于脑出血后7d开始增加,2周后同侧脑室下区(SVZ)及基底节区可见大量DCX阳性细胞,其表达达到高峰,为对照组的(793±192)%(P〈0.01)。结论成年大鼠中SVZ的神经元前体细胞在脑出血后明显增生,并向受损伤区域移行。
Objective To investigate neurogenesis in adult rat after intracerebral hemorrhage. Method Male Sprague-Dawley rats received an intracerebral infusion of 100μl autologous blood. Western blot analysis and histological examination were performed to check doublecortin ( DCX ) and Bromodeoxyuridine(Brdu). Results DCX levels in the ipsilateral basal ganglia started to increase as early as seven days after intracerebral hemorrhage ( ICH), peaked at 14 days (793±192 % of sham, P 〈 0.01 ). We found that most DCX positive cells were Brdu positive. Conclusion The results demonstrated that neurogenesis occurs from subventricular zone(SVZ) in the brain following ICH.
