摘要 目的探讨CA125阴性卵巢上皮性肿瘤患者血清相关蛋白及意义。方法取CA125阴性卵巢上皮性肿瘤和卵巢良性肿瘤、盆腔良性病变和正常妇女血清,采用双向电泳技术分离和鉴定生理状态与病理状态的差异表达蛋白,对有差异性表达的蛋白质运用MALDI-TOF-TOF串联质谱分析。结果成功获得重复性好的双向电泳图谱,CA125阴性卵巢上皮性肿瘤患者血清蛋白质表达谱发生了改变,综合分析得到CA125阴性时的蛋白质有8种,分别为结合珠蛋白α1、prostasin、calgranulin A、calgranulin B、溶血磷脂酸、增殖细胞核抗原、HSP60和HSP90。结论双向电泳—质谱法对卵巢上皮性肿瘤患者血清蛋白获得重复性好的结果,上述8种蛋白能为卵巢上皮性肿瘤提供早期诊断指标。 Objective To explore the significance of the serum associated protein in patients with CA125 negative ovarian epithelial tumor. Methods Serum was collected in patients with CA125 negative epithelial tumor of ovary, with benign tumor, with benign lesion and in healthy women. Separating and identifying the differential expression protein in physiological functions and pathologic state by two-dimensional electrophoresis, and the differential expression protein was analyzed with MALDI-TOF-TOF. Results The expression pattern in the serum in patients with CA125 negative epithelial tumor of ovary was changed with 2DE. 8 kinds of protein in patients with CA125 negative epithelial tumor of ovary were obtained. They were Hpα1, prostasin, calgranulin A and calgranulin B, LPA, PCNA, HSP60 and HSP90. Conclusion 2- DE-MS is stabe in detecting serum protein in patients with CA125 negative epithelial tumor of ovary. The above 8 proteins may be used to diagnose ovarian epithelial tumor early.
