摘要
本文采用丹磺酰肼荧光标记甾类化合物和反相高效液相色谱分离的方法,测定了多种甾类化合物包括甾体激素。通过实验优化了化学衍生反应的条件,反应的产率相当稳定,检出极限达2—3pmol,相对标准偏差1.46%,适于进行痕量分析。
A method of reversed-phase HPLC separation and highly sensitive fluorescence detection has been developed to determine dansyl hydrazine derivatives of some steroids including steroid hormones. After a simple step of derivatization at 60℃ in the presence of trifluorcacetic acid, the derivatives were chromatographed on a C_(18) column with 68% acetonitrile and 32% water (v/v) as the eluant. Optimization of derivatization reaction has been studied. The limit of detection is 2—3 pmol and relative standard deviation is 1.46%. Experimental results show that the method is reliable and accurate for trace analysis of steroids.
